Pharmasset is developing 2 polymerase inhibitors R7128 and this newer one
PSI-7851. We can look forward to the future of HCV therapy which will be
needed to have the most effective therapy and that will be oral HCV therapy
regimens that include at least 2 orally administered drugs. For the time
being peginterferon and ribavirin will have to be included in a therapy
regimen, but researchers hope to be able to eliminate peg/RBV from therapy but
this will have to be studied and they will have to prove in the studies that
peg and RBV can be eliminated from therapy, and whether we can or not get
rid of peg/RBV has some controversy as many researchers if not most feel
they can eliminate peg/RBV which will mean that for most patients the most
effective regimen will be 3 or 4 orally administered drugs. Perhaps they will
discover that the pegIFN dose can be reduced if used along with an oral
regimen. In addition there are 2 new interferons that might be useful if we
cannot eliminate peginterferon. Remember HCV is curable with time-limited
therapy so peg/RBV might be required to achieve the elimination of HCV without
ongoing therapy. Lambda interferon appears in early study to have little or
none of the typical interferon side effects. The first larger study in
patients was presented at EASL in April 2009, and a second interferon is from
Biolex called Locterin and this interferon has side effects but they are
less than with standard peginterferon. the third interferon which just
completed phase 3 is Albuferon which is admistered every 2 weeks but the side
effect profile did not look that different or better in phase 3 compared to
standard peginterferon, so the company developing it Human Genome Sciences
and Novartis has rights will have to establish with a new analysis that
taking Albuferon every 2 weeks results in better tolerability. Because the phase
3 studies presented at EASL showed a similar side effects profile when
looking at the numbers and types of side effects. Here are links to the EASL
